Description

From Solis BioDyne, this hot-start PCR reagent system belongs to the Hot-start PCR Enzymes and Master Mixes (5x), Standard cycling category [C0]. It is designed for amplifying difficult GC-rich templates and templates with stable secondary structures, demonstrating strong amplification with targets up to 79% GC content [S3]. The kit centres on a 5x concentrated, ready-to-use Master Mix that contains the chemically modified HOT FIREPol® DNA Polymerase for hot-start activation, 5x HOT FIREPol® GC Buffer, dNTPs, and BSA, with 7.5 mM MgCl2 in the mix delivering 1.5 mM MgCl2 in the final 1x PCR solution [S3].

100% DMSO and 25 mM MgCl2 are supplied in separate vials, giving users flexibility to titrate additives and magnesium for challenging amplicons [S3]. Reaction setup can be performed at room temperature, and the reagents are shipped at ambient temperature [S3]. Under routine storage at –20°C (–28°C to –18°C) the product is stable until the expiry date; it also remains stable for 6 months at 4°C (2–8°C) or 1 month at 25°C, with freeze–thaw stability of 30 cycles [S3]. One enzyme unit is defined as the amount required to catalyse the incorporation of 10 nmol of dNTPs into an acid-insoluble form in 30 minutes at 74°C [S3].

Manufactured by Solis BioDyne OÜ under ISO 9001 and ISO 13485 certified quality management systems, the product is supplied for research use only, and FIREPol is a registered trademark of Solis BioDyne OÜ [S2][S3]. A recommended cycling protocol begins with 12 minutes at 95°C to activate the polymerase, followed by 25–30 cycles of 95°C denaturation, 54–66°C annealing, and 72°C extension [S3].
