HOT FIREPol® Blend Master Mix with 10 mM MgCl2

A 5x ready-to-use master mix from Solis BioDyne designed for hot-start PCR under standard cycling conditions. The blend combines two carefully optimized enzymes – HOT FIREPol® DNA polymerase and a proofreading polymerase – providing both 5' flap endonuclease activity and 3'→5' proofreading activity. Fidelity is increased up to five fold compared to HOT FIREPol® alone, and the generated PCR products are compatible with both blunt-end and TA cloning procedures.

The formulation supplies 10 mM MgCl2 in the 5x stock (2 mM in 1x PCR solution), 1 mM of each dNTP (200 µM each at 1x), and BSA. Solis BioDyne recommends setting up reactions at room temperature, and the product is suited to PCR applications that will be visualized by agarose gel electrophoresis and ethidium bromide staining. Suggested cycling conditions are: initial activation at 95°C for 12–15 min; denaturation at 95°C for 10–20 s; annealing at 54–66°C for 30–60 s; elongation at 72°C for 20 s to 4 min; and final elongation at 72°C for 5–10 min, over 25–30 cycles.
