HOT FIREPol® Blend Master Mix with 10 mM MgCl2

From Solis BioDyne, this 5x hot-start master mix is supplied as a premixed, ready-to-use solution containing all of the reagents required for PCR apart from template, primers and water. The blend brings together HOT FIREPol® DNA polymerase and a proofreading polymerase, giving the mix both 5' flap endonuclease and 3'→5' proofreading activity in a single formulation.

Compared with HOT FIREPol® used on its own, the Blend Master Mix delivers increased fidelity of up to five-fold, and the PCR products generated are compatible with blunt-end and TA cloning procedures. The mix is designed for hot-start PCR and is recommended for applications that will be visualised by agarose gel electrophoresis with ethidium bromide staining. Reaction setup at room temperature is recommended, ideally under a laminar flow hood or in a PCR box to guard against contamination.

The 5x formulation provides 10 mM MgCl2 (yielding 2 mM in a 1x PCR solution) together with 1 mM of each dNTP (200 µM dATP, dCTP, dGTP and dTTP in 1x) and BSA. Routine storage is at -20 °C, while shipping or temporary storage at room temperature for up to 1 month, or at 2–8 °C for up to 6 months, has no detrimental effect on product quality.
