HOT FIREPol® Blend Master Mix Ready to Load with 7.5 mM MgCl2

From Solis BioDyne, this ready-to-load hot-start PCR master mix is supplied as a 5x premixed solution containing all reagents required for PCR except template, primers and water. The formulation includes a compound that enables direct loading onto agarose gels, along with two tracking dyes: a blue dye migrating equivalent to a 3.5–4.5 kb DNA fragment and a yellow dye migrating equivalent to a 35–45 bp DNA fragment.

The blend combines two enzymes—HOT FIREPol® DNA polymerase and a proofreading polymerase—providing both 5' flap endonuclease and 3'→5' proofreading activities, with the product exhibiting an increased fidelity (up to five fold) compared to HOT FIREPol® alone. Generated PCR products are compatible with blunt-end and TA cloning procedures, and the mix is described as offering increased yield, sensitivity and specificity. The 5x master mix buffer contains 7.5 mM MgCl2 (yielding 1.5 mM MgCl2 in a 1x PCR solution), 1 mM of each dNTP (200 µM each of dATP, dCTP, dGTP and dTTP in the 1x solution), and BSA.

Routine storage is recommended at -20ºC; shipping or temporary storage for up to 1 month at room temperature, or up to 6 months at 2–8ºC, has no detrimental effects on quality. Reaction setup at room temperature is highly recommended. The product is intended for hot-start PCR applications, is labelled for in vitro use only, and is supplied for research use only.
