Description

This cDNA synthesis kit from Solis BioDyne features a genetically modified MMLV-based reverse transcriptase with full RNase H activity, stabilised by a patented genetic modification referred to as the Stability TAG. The enzyme operates across a temperature range of 37°C – 60°C, with an optimum at 50°C, and is able to transcribe up to 8.9 kb RNA sequences with reaction times of 5–30 minutes. It supports oligo(dT), random, and gene-specific priming options and is suited to applications including gene expression analysis, disease diagnosis, genomic analysis, and structural studies of mRNA.

Supplied components include FIREScript® Reverse Transcriptase at 200 U/µl and 10x RT Reaction Buffer with DTT. The reverse transcriptase is purified from an E. coli strain carrying an overproducing plasmid containing the FIREScript Reverse Transcriptase gene. One unit is defined as the amount of enzyme that incorporates 1 nmol of dTTP into acid-precipitable material in 10 minutes at 37°C using poly(rA)•oligo(dT) as a template in a total reaction volume of 50 µl. The recommended reaction setup is performed at room temperature; for random or mixed primers, primer extension is carried out at 25°C for 5–10 minutes, reverse transcription at 50°C for 5 minutes, and enzyme inactivation at 85°C for 5 minutes.

The kit is shipped at room temperature and should be stored at –20°C (–28°C to –18°C) upon receipt until the expiry date. The enzyme remains stable at 4°C (2°C to 8°C) for 6 months, at room temperature (15–25°C) for 1 month, and tolerates 30 freeze-thaw cycles. It is manufactured by Solis BioDyne OÜ under an ISO 9001 and ISO 13485 certified quality management system and is supplied for research use only.
