FIREPol® Master Mix Ready to Load (7.5 mM MgCl2)

A 5x-concentrated ready-to-use solution from Solis BioDyne supplies all reagents required for PCR — except template, primers and water — together with two tracking dyes (blue and yellow) that allow progress to be monitored during electrophoresis. The mix contains FIREPol® DNA polymerase, 5x Reaction Buffer B (0.4 M Tris-HCl, 0.1 M (NH4)2SO4, 0.1% w/v Tween-20), 7.5 mM MgCl2 (yielding 1.5 mM MgCl2 in 1x PCR solution) and 1 mM of each dNTP (yielding 200 µM of dATP, dCTP, dGTP and dTTP in 1x PCR solution). An additional compound increases sample density for direct loading onto agarose gels.

The blue dye migrates equivalent to 3.5–4.5 kb DNA fragments, and the yellow dye migrates equivalent to 35–45 bp DNA fragments. The master mix is suited for a wide range of PCR assays and is recommended for applications visualised by agarose gel electrophoresis and ethidium bromide staining, but is not recommended where spectrophotometric measurements (absorbance or fluorescence) are required, as the dyes can interfere. Routine storage is at -18°C to -28°C, and shipping or temporary storage for up to 1 month at room temperature, or up to 6 months at 2–8°C, has no detrimental effects on the quality of the product.
