STEMdiff Microglia Differentiation Kit

From STEMCELL Technologies, the STEMdiff Microglia Differentiation Kit is part of a two-stage culture system used together with the STEMdiff Microglia Maturation Kit to generate microglia from human pluripotent stem cell (hPSC)-derived hematopoietic progenitor cells. The workflow begins with hematopoietic progenitor cells produced using the companion STEMdiff Hematopoietic Kit, after which the cells are cultured in differentiation medium and then transitioned to maturation medium.

The differentiation kit includes Microglia Basal Medium (90 mL, stored at 2 - 8 °C), Microglia Supplement 1 (10 mL, stored at -20 °C), and Microglia Supplement 2 (400 μL, stored at -20 °C); all components are stable for 2 years from the date of manufacture. Complete differentiation medium (Basal Medium plus Supplements 1 and 2) can be stored at 2 - 8 °C for up to 4 weeks after preparation. The resulting cell population is described as highly pure, with greater than 80% CD45/CD11b-positive cells, greater than 50% TREM2-positive microglia, and less than 20% morphologically distinct monocytes or macrophages.

Cells generated using the system are described as versatile tools for modeling human neurological development and disease, drug screening, toxicity testing, and cell therapy validation.
