STEMCELL Technologies offers a reagent kit for the immunomagnetic negative isolation of untouched human B cells from whole blood, spleen, or lymph nodes, using an immunodensity negative selection cocktail and column-free immunomagnetic separation. The workflow runs on a fully automated cell separation instrument that can perform up to four simultaneous or sequential separations, producing fractions ready for serology or flow cytometry crossmatch assays.

Starting with human whole blood from normal healthy donors, the B cell (CD19+) content of the non-lysed final isolated fraction typically ranges from 89 - 99.8% (gated on CD45); in a cited example the lysed whole blood start sample was 12.3% and the non-lysed final isolated fraction was 98.4% (gated on CD45). The kit incorporates mouse monoclonal antibodies against CD19 (human, chimpanzee), CD20 (human, rhesus, cynomolgus), and CD45 (human, chimpanzee) to support the negative selection of B cells.
