STEMdiff Gastric Organoid Diff Kit

A serum-free cell culture medium system from STEMCELL Technologies that enables robust and efficient generation of human pluripotent stem cell (hPSC)-derived gastric organoids through a four-stage differentiation protocol. hPSCs are directed through induction of definitive endoderm, posterior foregut spheroids, stomach patterning, and generation of gastric organoids. Differentiated cells form organoids composed of gastric epithelia and a surrounding, niche factor-producing mesenchyme, with epithelial and mesenchymal components expressing key markers of these gastric compartments. Organoids can be fully differentiated or maintained long-term through passaging or cryopreservation.

The kit supports two workflows: differentiation directly from hPSCs, based on the protocol published by McCracken et al., and long-term expansion and cryopreservation of gastric organoids, based on the formulation described by Bartfeld et al. The medium has been optimized to increase efficiency and reproducibility of organoid formation and expansion across hPSC lines, and is intended for differentiation of cells maintained in mTeSR™1 or mTeSR™ Plus. Applications include studying gastric development and cell biology, gastric inflammation and other pathologies, gastric regeneration, microbial interaction, disease modeling, drug discovery, and compound screening.
