For researchers building in vitro models of human cortical development, this forebrain neuron workflow from STEMCELL Technologies delivers a defined, serum-free system for converting hESCs and iPSCs into excitatory cortical-like neurons. The protocol drives cells through neural induction and forebrain-specific patterning, producing populations enriched for neurons expressing FOXG1, TBR1, and CTIP2 within roughly 10 to 14 days. The xeno-free formulation removes animal-derived components, lowering batch-to-batch variability and supporting reproducible in vitro disease modeling.

Typical applications include modeling neurodevelopmental and neurodegenerative disorders such as Alzheimer's, Parkinson's, schizophrenia, and autism spectrum conditions. The generated neurons support patch-clamp electrophysiology, calcium imaging, multi-electrode array recordings, high-content screening, and synaptic puncta analysis, and are widely used for neurotoxicity screening and patient-derived iPSC drug discovery programs.

Each kit is supplied as a basal medium paired with frozen forebrain patterning supplements in single-use vials. Supplements are thawed, aliquoted, and held at -20°C to survive repeated freeze-thaw use, while the basal medium is stored refrigerated and protected from light. The workflow runs in either monolayer or embryoid body starting cultures; a typical plating volume is around 50 uL of cell suspension onto poly-L-ornithine and laminin-coated wells. Components are lot-traced and intended for research use only.
