STEMdiff Choroid Plexus Org Mat Kit

The STEMCELL Technologies STEMdiff™ Choroid Plexus Organoid Maturation Kit is a culture medium kit for the establishment of organoid models containing human pluripotent stem cell-derived choroid plexus-like epithelium. Designed to further develop hPSC-derived neuroepithelial tissues for CNS permeability and CSF biomarker assays, the kit is used in a 30-day protocol that begins with embryoid body formation, followed by expansion of neuroepithelia and patterning to choroid plexus-like epithelium.

As part of the workflow, the maturation medium is applied from day 15, with feeding every 2 - 3 days. After a period of epithelial maturation including extensive bubbling, organoids develop cystic structures surrounded by an ependymal epithelial layer and filled with a fluid resembling cerebrospinal fluid (CSF). At day 40, organoids display translucent cysts containing CSF-like fluid, with epithelia exhibiting the tightly packed and cuboidal morphology typical of ependymal cells that line the brain ventricles in vivo. Immunohistological analysis shows that the cystic structures are TTR- and CLIC6-positive, with these ependymal cell markers highly co-localized.

CSF-like fluid extracted from organoid cysts has been shown by western blot analyses to contain Clusterin and IGF-2 proteins. Organoids can be generated from human embryonic stem (ES) or induced pluripotent stem (iPS) cells.
