STEMdiff Cardiomyocyte Expansion Kit

STEMCELL Technologies offers this serum-free kit for the expansion of early-stage human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs). Designed for use in disease modeling, toxicology, and cell therapy research, the kit supports expansion of hPSC-CMs that have been differentiated using the STEMdiff Ventricular or Atrial Cardiomyocyte Differentiation Kit. Expanding cells are characterized by the cardiac muscle cell marker cardiac troponin T (cTnT) and can be passaged up to 5 times. The kit is used alongside the STEMdiff Cardiomyocyte Support Medium and STEMdiff Cardiomyocyte Dissociation Kit to support passaging and dissociation workflows, and expanded cultures can be transitioned to the STEMdiff Cardiomyocyte Maintenance Kit for long-term maintenance or downstream assays.

Performance data from studies across 4 hPSC lines show that, after 5 passages, cardiomyocyte cultures achieve an average cTnT purity of 90% ± 2% (n = 15), with cumulative fold expansion ranging from 33- to 103-fold and averaging 72-fold ± 12 (n = 15, 4 hPSC lines). Post-expanded hPSC-derived cardiomyocytes replated onto microelectrode arrays and maintained for 1 week exhibit a stable beat rate averaging 12 ± 1 beats per minute (n = 14, 4 hPSC lines).
