STEMdiff BLO Maturation Kit

A serum-free medium system from STEMCELL Technologies that supplies the components required for the maturation phase of branching lung organoid cultures generated from human ES and iPS cells. The kit is intended for use during extended organoid culture beyond 28 days in a Matrigel® sandwich.

When applied to long-term cultures, branching tips of branching lung organoids continue to grow and branch up to day 101, and expression levels of more mature distal lung markers ABCA3, SFTPC, and SFTPB increase over time, as assessed by RT-qPCR with RQ-values normalized to TBP and compared to commercially available lung RNA.

The kit includes a lung basal medium, a 10X lung supplement containing material derived from human plasma (donors have been tested and found negative for HIV-1, HIV-2, hepatitis B, and hepatitis C prior to donation), and a 100X branching lung organoid Supplement B that is light sensitive and should be handled with minimized light exposure.
