STEMdiff Astrocyte Differentiation Kit

A differentiation kit from STEMCELL Technologies is designed to rapidly and efficiently generate astrocytic precursors from neural progenitor cells (NPCs) derived from human pluripotent stem cells (hPSCs). According to the manufacturer product information sheet, the workflow begins with NPCs produced using the STEMdiff™ SMADi Neural Induction Kit, and the resulting astrocytic precursors are subsequently matured into astrocytes using the separately sold STEMdiff™ Astrocyte Maturation Kit. Using this system, a highly pure population of astrocytes—more than 70% S100β-positive and more than 60% GFAP-positive, with fewer than 15% doublecortin-positive neurons—can be generated from hPSCs in as little as 7 weeks, and the cells can be maintained long-term in culture. Cells derived with this workflow are described as versatile tools for modeling human neurological development and disease, drug screening, toxicity testing, and cell therapy validation. The kit is also referenced in a manufacturer protocol for establishing a two-dimensional tri-culture of hPSC-derived forebrain neurons, astrocytes, and microglia to support studies of neuronal–glial and neuroimmune interactions, and is noted for allowing experimental flexibility through customization with specific genotypes of interest.
