Description

From Solis BioDyne, this kit is an optimized qPCR solution compatible with lyophilization, designed for fast and sensitive DNA amplification with probe-based assays. The formulation includes a glycerol-free qPCR mix and a lyophilization excipient mix, suitable for reliable lyophilization into cakes and beads. The critical temperature (Tc) of the kit is -30.8 °C and the glass transition temperature (Tg) of the lyophilized product is 68.3 °C.

The 5x SolisFAST® Lyo-compatible qPCR Mix with UNG is a 5x concentrated glycerol-free master mix supporting real-time detection and quantitation of up to five DNA targets simultaneously. It contains the SolisFAST® DNA polymerase, an in silico designed inhibitor tolerant analogue of Taq DNA polymerase with oligo-based hot-start and ~2-4 times faster extension rates compared to wild-type Taq. The mix includes dUTPs in place of dTTPs together with Salini UNG® Uracil-N-Glycosylase to prevent carry-over contamination. The MgCl2 concentration is 16.5 mM (3.3 mM at 1x). The 4x SolisFAST® Lyo Excipient Mix is a concentrated blend of additives that protect reagents during freeze-drying and stabilize the lyophilizates, and is compatible with qPCR cyclers that do not require an internal reference dye such as ROX.

Storage options: routine storage at -20 °C until expiry, stable at 4 °C for 5 months, stable at room temperature (25 °C) for 2 weeks, with freeze-thaw stability for 30 cycles. Reaction setup is performed at room temperature. The product is manufactured in compliance with ISO 9001 and ISO 13485. Recommended cycling: 3 min initial denaturation at 95 °C, followed by 40 cycles of denaturation at 95 °C and annealing/extension at 60-65 °C. A final concentration of 400 nM each primer and 150 nM probe is suitable for most applications. For reconstitution, the same volume of DNA sample as the total pre-lyophilization reaction volume is recommended (e.g., 20 µl of DNA sample for a 20 µl reaction). The product is supplied for research use only.
