SolisFAST® 1-step RT-PCR Kit with UNG

From Solis BioDyne, this kit is designed for sensitive and highly specific cDNA synthesis and end-point PCR in a single tube, combining an exceptionally fast and robust SolisFAST® polymerase with highly processive reverse transcriptases. The format delivers a streamlined workflow for analysing RNA targets by reverse transcription PCR, with cDNA synthesis and PCR performed in one tube to reduce reaction-to-reaction variation, minimise hands-on steps and shorten time to results. The product is supplied as a kit of four tubes containing all components required to perform RT-PCR except RNA template and primers.

The 4x One-step HOT SolisFAST® Mix with UNG includes HOT SolisFAST® DNA Polymerase, a chemical hot-start in silico designed analogue of Taq DNA polymerase with approximately 2–4 times faster extension rates, together with an optimised buffer, dNTPs including dUTP, and heat-inactivatable Salini UNG® Uracil-N-Glycosylase to prevent carry-over contamination. The 40x One-step RT Mix contains the thermostable SOLIScript® and FIREScript® Reverse Transcriptases, active at temperatures up to 60°C and useful with templates containing high levels of secondary structure, together with RiboGrip® RNase Inhibitor, which inactivates commonly encountered RNases (A, B and C). A separate 5x AmpliBoost™ RT-PCR Enhancer is included to improve sensitivity and performance with GC-rich and other challenging amplicons.

Optimised composition enables detection of as little as 0.1 pg of RNA and supports multiplexing of up to 5 RNA targets simultaneously, making the kit suitable for virus detection as well as for both singleplex and multiplex RT-PCR assays. The recommended total RNA input ranges from 0.1 pg to 1 µg in a 20 µl reaction volume. The product is inherently stable due to proprietary Stability TAG technology and is manufactured under an ISO 9001 and ISO 13485 certified Quality Management System. For routine use the kit is stored at −20°C until the expiry date, with a stated freeze-thaw stability of 10 cycles.
