Description

A thermostable hot-start DNA polymerase from Solis BioDyne, designed for MALDI-TOF mass spectrometry and other primer extension platforms. The enzyme is chemically modified and remains inactive at ambient temperature, with no polymerase activity until fully activated by a 12–15 minute incubation at 95°C, a feature that prevents the extension of non-specifically annealed primers.

The polymerase exhibits 5'→3' polymerase activity and shows enhanced efficiency for incorporating unconventional nucleotides, including ddNTPs and labeled ddNTPs. Beyond MALDI-TOF, the enzyme is suited to primer extension and Mass-array workflows. It is supplied at a concentration of 5 U/µl, with an error rate per nucleotide per cycle of approximately 8 × 10⁻⁵ and an estimated half-life at 95°C of 1.5 hours. One unit is defined as the amount of enzyme required to catalyze the incorporation of 10 nmol of dNTP into an acid-insoluble form in 30 minutes at 74°C.

The product is purified from an E. coli strain carrying an overproducing plasmid that contains a modified gene of Thermus aquaticus DNA Polymerase. The storage and dilution buffer contains 50% glycerol (v/v), 20 mM Tris-HCl pH 8.7 at 25°C, 100 mM KCl, 0.1 mM EDTA, and stabilizers. The accompanying 10x Reaction Buffer C is composed of 500 mM Tris-HCl pH 9.5 at 25°C and 100 mM MgCl₂. Routine storage is at −20°C and the product is shipped at room temperature. For research use only.
