SepMate-50 100-pack RUO

Manufactured by STEMCELL Technologies, this research-use-only SepMate-50 tube system is designed to isolate mononuclear cells (MNCs), comprising lymphocytes and monocytes, from whole blood or bone marrow by density centrifugation. The tube features a specialized insert that minimizes mixing of the sample and the density gradient medium, removing the need for careful layering and careful cell removal from the interface. Density gradient medium is pipetted through a central hole in the insert, partially filling the tube, after which whole blood is rapidly pipetted down the side of the tube to rest upon the medium.

After centrifugation at 1200 x g for 10 minutes at room temperature with the brake on, the enriched cell layer is poured off into a new tube, while density gradient medium, erythrocytes, and granulocytes are retained below the insert. For samples older than 24 hours, a centrifugation time of 20 minutes is recommended. The MNCs are then washed and are ready for use. The SepMate-50 is suitable for processing 4 - 17 mL of initial sample with a minimum packed RBC volume of 2 mL and a maximum packed RBC volume of 12 mL, requiring 15 mL of density gradient medium. The product is compatible with human whole peripheral blood, bone marrow, and cord blood samples, and is not intended for use with leukapheresis samples, buffy coat samples, or samples older than 48 hours. It can also be used with RosetteSep cell enrichment cocktails to isolate specific cell types from human whole blood.

Tubes are sterilized by an electron beam process conforming to ISO 11137-1, and should be stored at ambient temperature until the expiry date on the label. They are for single use only and should be handled by trained personnel observing good laboratory practice.
