Description

From STEMCELL Technologies, the RoboSep™ Release Human CD45 Positive Selection Kit for Humanized Mice uses immunomagnetic positive selection with particle release technology to isolate highly purified and magnetic particle-free human CD45+ leukocytes. Designed for single-cell suspensions of primary human tissues and tumors, as well as tissues and tumor xenografts from humanized mice, the kit combines monoclonal antibody specificity with a column-free magnetic system. EasySep™ has been used in published research for more than 20 years, and highly purified CD45+ cells can be isolated in less than 45 minutes.

Cells are labeled with antibody complexes recognizing CD45 along with magnetic particles called EasySep™ Releasable RapidSpheres™. After separation using an EasySep™ magnet, the bound magnetic particles are released from the isolated CD45+ cells using a release agent. The final fraction contains highly purified CD45+ cells ready for downstream applications including flow cytometry, culture, and DNA/RNA extraction. The selection antibody is a mouse monoclonal IgG1 against human and chimpanzee CD45, and following isolation these antibody complexes remain bound to the cell surface and may interact with Brilliant Violet™ antibody conjugates, polyethylene glycol-modified proteins, or other chemically related ligands.

Manufacturer data show that starting with a single-cell suspension of a human breast cancer tumor xenograft sample from an NRG-3GS humanized mouse, the CD45+ TIL purities of the starting and final isolated fractions were 5.5% and 96.0%, respectively. Mass cytometry analysis with 19 markers confirmed that EasySep™-isolated CD45+ cells are representative of the starting leukocyte population. In functional assays, pre- and post-isolation PBMCs incubated for 24 hours with peptide pools or the mitogen PHA produced IFN-gamma in ELISpot assays. The kit has been used to isolate highly purified CD45+ cells from spleens, lungs, bone marrow, and tumors in humanized mouse xenograft models.
