RosetteSep Monocyte CD36 Depletion, 2mL

From STEMCELL Technologies, this immunorosetting cocktail is designed to deplete human CD36+ monocytes directly from whole blood, leaving behind an enriched fraction of untouched, viable cells that are immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction. The product is intended to process up to 40 mL of whole blood and is described as fast and easy-to-use, requiring no special equipment or training. It can be combined with SepMate™ tubes for consistent, high-throughput sample processing.

The antibody cocktail crosslinks unwanted monocytes in whole blood to multiple red blood cells (RBCs), forming immunorosettes that increase the density of the targeted cells so they pellet with free RBCs during centrifugation over a density gradient medium. Desired cells are collected as a highly enriched population at the interface between the plasma and the density gradient medium. For optimal performance, whole peripheral blood should be used within 24 hours of collection and stored at room temperature (15 – 25 °C), with a nucleated cell concentration not exceeding 5 × 10⁷ cells/mL. The cocktail is a combination of monoclonal antibodies in phosphate-buffered saline (PBS).

The cocktail should be stored at 2 – 8 °C and not frozen, and components may be shipped at 15 – 25 °C. The recommended medium is Dulbecco's Phosphate Buffered Saline with 2% Fetal Bovine Serum, and the density gradient medium should have a density of 1.077 g/mL. In an example experiment described in the product documentation, purities of the start and final depleted fractions were 6% and 0.04%, respectively, representing 2.8 Log depletion.
