Description

This immunodensity-based cell isolation reagent from STEMCELL Technologies is designed to deplete human monocytes directly from whole blood. The cocktail contains a combination of monoclonal antibodies that crosslink unwanted monocytes to multiple red blood cells through tetrameric antibody complexes recognizing CD36 and glycophorin A, forming immunorosettes. When centrifuged over a buoyant density medium, the increased density of these rosettes causes them to pellet along with free RBCs, leaving desired cells as a highly enriched population at the interface between plasma and the density gradient medium. The untouched, viable cells are immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction.

The product is fast and easy to use, requires no special equipment or training, and can be combined with SepMate™ tubes for consistent, high-throughput sample processing. The cocktail should be stored at 2 - 8°C (do not freeze) and is stable until the expiry date on the label; it may be shipped at 15 - 25°C. A precipitate may be observed in the vial but will not affect performance.

For optimal performance, whole peripheral blood should be used within the last 24 hours and kept at 15 - 25°C, and the concentration of nucleated cells should not exceed 5 x 10^7 cells/mL. The recommended medium is Dulbecco's Phosphate Buffered Saline with 2% fetal bovine serum, and the recommended density gradient medium has a density of 1.077 g/mL. For purity assessment by flow cytometry, anti-human CD14 antibody clones M5E2 or MoP9 are suggested. In a representative example, the purities of the start and final depleted fractions were 6% and 0.04%, respectively, representing 2.8 Log depletion. The product is part of the RosetteSep™ portfolio for immunology research.
