The 0.5 mL vial from STEMCELL Technologies contains a tetrameric antibody complex cocktail for the negative selection and enrichment of human NK cells directly from whole blood. After a 20-minute room temperature incubation at 50 µL per mL of whole blood, unwanted leukocytes crosslink to red blood cells via tetrameric complexes and sediment during standard Ficoll-Paque density gradient centrifugation, allowing NK cells to be harvested from the plasma interface as an unlabeled, viable population. The aqueous cocktail ships frozen on dry ice and should be stored frozen at -20°C to preserve activity. The enrichment procedure is compatible with downstream flow cytometry, cytotoxicity assays, and in vitro cell culture applications. Each 0.5 mL vial processes up to 50 mL of human whole blood, with typical NK cell purity reaching 70-90% depending on donor variability. Human-specific antibodies minimize off-target reactivity, producing a population ready for functional immunology studies without the need for magnetic columns or bead-based separation.
