Negative-Selection Monocyte Enrichment from Whole Blood

This 0.5 mL RosetteSep reagent from STEMCELL Technologies isolates CD14+ CD16+ monocytes directly from human whole blood without columns, magnets, or positive selection. The cocktail contains tetrameric antibody complexes that simultaneously bind unwanted leukocytes (T cells, B cells, NK cells, and granulocytes) and glycophorin A on red blood cells, forming dense rosettes that pellet through a density gradient medium during centrifugation. The monocyte fraction collects at the plasma interface as an untouched population, retaining native surface marker expression and functional responsiveness for downstream characterization or culture.

For each 0.5 mL unit, approximately 5 mL of fresh whole blood collected in EDTA or heparin is processed. After a 20 minute room temperature incubation with the reagent, the sample is layered over Lymphoprep or an equivalent density gradient medium and centrifuged briefly. Isolated cells can be washed, counted, and used immediately for flow cytometry panels, cell culture including macrophage differentiation, phagocytosis assays, monocyte-derived dendritic cell work, cytokine profiling, or downstream RNA and DNA extraction workflows. Researchers commonly apply this approach to studies of autoimmune disease, infectious disease, atherosclerosis, transplant immunology, and cancer research where primary monocyte responses are the focus.

Supplied as a single vial of ready-to-use cocktail; the 0.5 mL format is intended as a single-use aliquot for small-volume studies. Refrigerate upon receipt and protect from light; do not freeze. For in vitro research use only, not for diagnostic or therapeutic applications. Each lot is verified for expected monocyte recovery and viability on human whole blood donors.
