Description

This immunorosette-based cell depletion reagent from STEMCELL Technologies is designed to remove CD8+ cells directly from human whole blood, yielding an untouched, viable population that is immediately ready for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction. According to the product information sheet, this size is configured for processing up to 40 mL of whole blood. The antibody cocktail targets CD8+ cells for removal with antibodies recognizing CD8 and glycophorin A surface markers, crosslinks unwanted cells to multiple red blood cells (RBCs) to form immunorosettes; these dense rosettes pellet together with free RBCs during centrifugation over a density gradient medium, while the desired cells remain unlabeled and are easily collected as a highly enriched population at the plasma–density gradient interface.

Fast and easy to use, the cocktail requires no special equipment or training and can be combined with SepMate™ tubes for consistent, high-throughput sample processing. It is supplied as a combination of monoclonal antibodies in PBS and should be stored at 2–8°C (do not freeze); components may be shipped at room temperature (15–25°C) and remain stable until the expiry date on the label. For optimal performance, whole peripheral blood should be used within 24 hours of collection and kept at room temperature (15–25°C). The cocktail is added at 50 µL per mL of sample, incubated at room temperature for 20 minutes (standard tube) or 10 minutes (SepMate tube), and centrifuged at 1200 × g. In the manufacturer's representative dataset, the purities of the start and final depleted fractions are 13% and 0.4%, respectively, corresponding to a 2.0 Log depletion. The recommended medium is Dulbecco's PBS containing 2% fetal bovine serum, and the protocol requires a density gradient medium with a density of 1.077 g/mL (such as Lymphoprep™).
