Description

An immunodensity negative selection cocktail from STEMCELL Technologies enriches untouched and highly purified circulating epithelial tumor cells (CTCs) directly from human whole blood by negative selection. The antibody cocktail crosslinks unwanted cells to multiple red blood cells, forming immunorosettes that pellet along with free RBCs during density gradient centrifugation, leaving desired cells unlabeled and easily collected as a highly enriched population at the plasma–density gradient medium interface.

Designed to be fast and easy-to-use without columns, the product can be combined with SepMate™ tubes for consistent high-throughput sample processing, achieving up to 98% purity. The cocktail contains a combination of mouse and rat monoclonal antibodies bound in bispecific Tetrameric Antibody Complexes (TACs) directed against cell surface antigens on human hematopoietic cells (CD3, CD14, CD16, CD19, CD38, CD45, CD56, CD61, and CD66b) and glycophorin A on RBCs; the mouse monoclonal antibody subclass is IgG1. Isolated cells are immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction, and purity can be assessed by staining with a fluorochrome-conjugated tumor-specific antibody.

For optimal performance, use whole peripheral blood collected within the last 24 hours and stored at room temperature (15 – 25°C). The cocktail is supplied as a combination of monoclonal antibodies in PBS, is stored refrigerated at 2 – 8°C, must not be frozen, and is stable until the expiry date on the label; it may be shipped at room temperature (15 – 25°C), contents have been sterility tested, and a precipitate may be observed but will not affect performance. According to manufacturer data, typical depletion of targeted CD45+ cells ranges from 3.4 to 4.7 log.
