Description

This immunodensity negative selection cocktail from STEMCELL Technologies enriches highly purified circulating tumor cells (CTCs) directly from human whole blood, achieving up to 98% purity with no columns required. Unwanted cells are crosslinked to multiple red blood cells (RBCs) via tetrameric antibody complexes that recognize specific cell surface markers, forming immunorosettes that pellet through a density gradient medium during centrifugation. Desired cells are never labeled with antibody and are collected as a highly enriched population at the plasma–density gradient medium interface.

The cocktail contains a combination of monoclonal antibodies in phosphate-buffered saline, including an anti-CD36 antibody together with reagents directed against cell surface antigens on human hematopoietic cells (CD3, CD14, CD16, CD19, CD38, CD45, CD56, CD61, and CD66b) and glycophorin A on RBCs. It can be combined with SepMate™ tubes for consistent, high-throughput sample processing. For optimal performance, whole peripheral blood should be used within 24 hours of collection and stored at room temperature. The nucleated cell concentration should not exceed 5 x 10^7 cells/mL, and RBCs should be present at a ratio of at least 50 - 100 RBCs per nucleated cell.

The cocktail is added at 50 µL/mL of whole blood, incubated for 20 minutes at room temperature, and centrifuged at 1200 x g for 20 minutes with the brake off over a density gradient medium with a density of 1.077 g/mL. Isolated cells are immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction. The product is stored at 2 - 8°C, should not be frozen, is stable until the expiry date on the label, and is for research use only.
