RosetteSep HLA Total Lymph Cktl 10mL

An immunodensity reagent from STEMCELL Technologies enriches untouched, highly purified lymphocytes directly from human whole blood by negative selection, making it well suited for HLA laboratory workflows including crossmatch analysis, HLA serological typing, and chimerism analysis. The RosetteSep™ cocktail contains a combination of monoclonal antibodies in PBS that crosslink unwanted cells in human whole blood to multiple red blood cells, forming immunorosettes. These rosettes increase the density of the unwanted cells so they pellet along with free red blood cells during density gradient centrifugation, leaving desired lymphocytes untouched and easily collected as a highly enriched population at the plasma–density gradient interface. Isolated cells remain viable and immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction.

The procedure is fast and easy to use, requires no special equipment or training, and can be combined with SepMate™ for consistent, high-throughput sample processing. Typical purity of enriched total lymphocytes (T [CD3+] and B [CD19+] cells) is greater than 80%. CD3+CD16+ NK cells are removed during the procedure. For purity assessment by flow cytometry, the manufacturer recommends Anti-Human CD3 Antibody, Clone UCHT1 and Anti-Human CD19 Antibody, Clone HIB19. The cocktail should be stored at 2–8°C and not frozen; components may ship at 15–25°C but must be stored as directed. Each lot is sterility tested according to USP methods and quality-control performance tested in cell separation assays using human whole blood.

The kit is intended for research use only and not for human or animal diagnostic or therapeutic uses unless otherwise stated. A density gradient medium with a density of 1.077 g/mL (such as Lymphoprep™ or RosetteSep™ DM-L Density Medium) and a recommended medium of Dulbecco's Phosphate Buffered Saline with 2% Fetal Bovine Serum are required but not provided.
