RoboSepHLA ChimWB Lymphoid Pos Slctn Kit

A column-free immunomagnetic positive selection kit from STEMCELL Technologies is designed to isolate highly purified human CD3+ T lymphocytes and CD19+ B lymphocytes from fresh human whole blood, buffy coat, or an LRS cone. The kit supports chimerism analysis following HSC or bone marrow transplantation using EasySep™ technology, requiring no chromatography columns. CD3 antigen is expressed on T lymphocytes and NKT cells, while CD19 antigen is expressed on B cells and follicular dendritic cells.

Targeted cells are labeled with a tetramer antibody complex recognizing CD3 and CD19 along with dextran-coated magnetic particles. The reagent mixture also includes antibodies directed against human Fc receptors to minimize non-specific binding. Labeled cells are separated with the EasySep™ magnet, with desired cells retained in the tube and unwanted cells poured off. The kit is supplied with RoboSep™ buffer and RoboSep™ filter tips.

In a representative run, the purities of the start and final isolated fractions were 16.2% and 98.5%, respectively. The lymphoid cell content (CD3+CD19+) of the isolated fraction is typically 99.5 ± 0.2% when gated on CD45 (mean ± SD using 'The Big Easy' EasySep™ Magnet). The kit is intended for use with human buffy coat, peripheral blood, and whole blood samples and supports immunology research applications. Compatible magnets include 'The Big Easy' EasySep™ Magnet and RoboSep-S™.
