RoboSepHLA Chim WB CD33 Pos Slctn Kit

STEMCELL Technologies offers this immunomagnetic kit for isolating highly purified CD33+ myeloid cells from fresh human whole blood or buffy coat for HLA chimerism analysis. The kit uses positive selection with an antibody recognizing the CD33 surface marker and requires no columns. Cells are labeled with antibodies and magnetic particles and separated using an EasySep™ magnet; unwanted cells are poured off while desired cells remain in the tube. Isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction for lineage-specific chimerism analysis.

The kit supports manual processing with "The Big Easy" EasySep™ Magnet or EasyEights™, as well as fully automated processing with the RoboSep™ instrument. Sample volume range is 0.25 - 4.5 mL, and the kit is designed for processing 60 mL of whole blood. The purity of the isolated CD33+ fraction is typically 96 ± 2% (mean ± SD) using "The Big Easy" EasySep™ Magnet, with up to 99% purity achievable.

Components include an EasySep™ HLA Chimerism Whole Blood CD33 Positive Selection Cocktail (a combination of monoclonal antibodies in PBS, including an Fc receptor blocking antibody), EasySep™ Dextran RapidSpheres™ (a suspension of magnetic particles), and EasySep™ Red Blood Cell Lysis Buffer (10X concentrate). Recommended antibodies for assessing purity by flow cytometry include anti-human CD14 (clones M5E2 or MoP9) and anti-human CD66b (clone G10F5). The product is for research use only.
