RoboSep Rat IgM Depl Kit

Manufactured by STEMCELL Technologies, this immunomagnetic kit is designed to deplete IgM+ cells from rat splenocytes, lymph nodes, or whole blood by positive selection. The protocol is fast, easy-to-use, and column-free, with up to 90% purity (≤ 10% IgM+). Isolated cells are untouched and immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction.

The kit targets IgM+ cells for removal with antibodies recognizing specific cell surface markers. Unwanted cells are labeled with antibodies and magnetic particles, then separated without columns using an EasySep™ magnet, and the desired cells are simply poured off into a new tube. It is for processing 1 x 10^9 cells, and when using a single-cell suspension from other tissue types the kit may require optimization.

Components include an EasySep™ Rat IgM Depletion Cocktail (a combination of monoclonal antibodies in PBS and 0.1% BSA), EasySep™ Dextran RapidSpheres™ (a suspension of magnetic particles in water), and an Anti-Rat CD32 Blocker (a combination of monoclonal antibodies in PBS, 0.1% BSA, and < 0.1% sodium azide). All components should be stored at 2 - 8°C and not frozen; they remain stable until the expiry date on the label. The kit has been tested with Sprague Dawley and Wistar rat strains and is expected to be compatible with other strains. For purity assessment of residual IgM+ cells by flow cytometry, anti-rat IgM antibody and anti-rat CD45RA antibody are recommended. In data from splenocytes of a Sprague Dawley rat, the IgM content of the final isolated fraction is typically 8.0 ± 4.1%, with start and final depleted fraction purities of 55.7% and 8.7%, respectively (1.6 log depletion).
