RoboSep Rat CD4+ T Cell Isolation Kit

This immunomagnetic negative selection kit from STEMCELL Technologies uses RoboSep™ automation and isolates untouched, highly purified CD4+ T cells from single-cell suspensions of rat splenocytes, lymph node, or whole blood. Use with other tissue types may require optimization. Non-CD4+ T cells are targeted for removal with antibodies recognizing specific cell surface markers; unwanted cells are labeled with antibodies and magnetic particles, and separated without columns using an EasySep™ magnet. Desired cells are simply poured off into a new tube and are immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction.

Designed for processing 1 x 10^9 cells, the kit has been verified for use with Sprague Dawley and Wistar rat strains and is expected to also be compatible with other strains. Isolations can be completed in as little as 13 minutes. Up to 98% purity is achievable, and starting with rat splenocytes the CD4+ T cell content (CD3+CD4+) of the isolated fraction is typically 97.8 ± 0.8% (mean ± SD using the purple EasySep™ Magnet with the optional additional separation). An optional second round of separation can, on average, improve purity by up to 10% but may reduce recovery by 5–15%.
