RoboSep Rat B Cell Isolation Kit

A column-free immunomagnetic negative selection kit from STEMCELL Technologies for isolating untouched, highly purified B cells from single-cell suspensions of rat splenocytes, lymph nodes, or whole blood. The kit targets non-B cells for removal with antibodies recognizing specific cell surface markers, and unwanted cells labeled with antibodies and particles are separated using an EasySep™ magnet rather than columns, allowing the desired cells to be simply poured off into a new tube. Isolated cells are immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction.

The kit is compatible with several magnets, including the EasySep™ Magnet, "The Big Easy" EasySep™ Magnet, Easy50, EasyEights™, and the RoboSep™ instrument for fully automated processing. It has been verified for use with the Sprague Dawley rat strain and is expected to also be compatible with other strains. For purity assessment of B cells by flow cytometry, anti-rat CD45RA antibody and anti-rat CD3 antibody are recommended.

According to the manufacturer, the B cell content (CD45RA+CD3-) of the isolated fraction is typically 94.1 ± 3.8% (mean ± SD using the purple EasySep™ Magnet), with the purities of the start and final isolated fractions reported as 44.5% and 95.6% respectively. The kit is designed for processing 1 x 10^9 cells and achieves up to 95% purity.
