RoboSep Negative Human Monocyte, KIT

From STEMCELL Technologies, this immunomagnetic negative selection kit — compatible with both manual EasySep™ magnets and the fully automated RoboSep™ system — isolates untouched and highly purified CD14+CD16+ human monocytes from fresh or previously frozen peripheral blood mononuclear cells (PBMCs) or lysed leukapheresis samples. The workflow is column-free: non-monocytes are labeled with antibodies and magnetic particles and separated using an EasySep™ magnet, while desired cells are simply poured off into a new tube. Isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.

The kit is fast and easy to use and yields up to 89% purity of untouched, viable cells. Starting with freshly prepared PBMCs, the CD14+ cell content of the enriched fraction is 82.7 ± 6.7% (mean ± SD using the purple EasySep™ magnet), with start and final enriched fractions of 21.2% and 83.3% respectively. Slightly lower CD14+ cell purities may be obtained from samples that contain a large number of CD16+ cells.

Designed for processing 1 x 10^9 cells, the kit is supplied with an EasySep™ Human Monocyte Enrichment Cocktail w/o CD16 Depletion — a combination of monoclonal antibodies in PBS that includes an Fc receptor blocking antibody — together with EasySep™ D Magnetic Particles for Human Monocytes, a suspension of magnetic particles in TBS. Both components should be stored at 2 - 8°C and are stable until the expiry date (EXP) on the label; they should not be frozen. The product is intended for research use only.
