RoboSep Ms PE Positive Selection Kit II

STEMCELL Technologies offers an immunomagnetic positive selection kit for isolating highly purified cells labeled with PE (phycoerythrin)-conjugated antibodies from mouse splenocytes, bone marrow, or other single-cell suspensions. The procedure is fast and easy-to-use, and requires no columns. Labeled cells are separated using a magnet, with unwanted cells simply poured off while desired cells remain in the tube. Isolated cells are immediately available for downstream applications such as flow cytometry, cell culture, or cell-based assays.

The kit comprises an EasySep™ PE Selection Cocktail, an EasySep™ Mouse FcR Blocker, and EasySep™ Dextran RapidSpheres™. For spleen samples, the tissue is disrupted in PBS or Hanks' Balanced Salt Solution containing 2% fetal bovine serum, passed through a 70 µm mesh nylon strainer, and resuspended at 1 x 10^8 nucleated cells/mL after centrifugation at 300 x g for 10 minutes. For bone marrow, cells are flushed from femur and tibia using a syringe equipped with a 23 gauge needle, then dispersed and processed through a 70 µm mesh nylon strainer.

In an example separation starting with mouse splenocytes, purities of 20.5% in the starting fraction and 91.6% in the final isolated fraction were obtained using a PE-conjugated anti-mouse CD4 antibody. A Product Information Sheet (version 03) is available from the manufacturer.
