RoboSep Mouse Naïve CD8+ T Cell Iso Kit

From STEMCELL Technologies, this immunomagnetic isolation kit enables researchers to obtain untouched, highly purified naïve CD8+ T cells (CD3+CD8+CD44-CD62L+) from mouse splenocytes or lymph nodes using a column-free negative selection workflow. Non-naïve CD8+ T cells are targeted for removal with biotinylated antibodies, and unwanted cells labeled with streptavidin-coated magnetic particles are separated using an EasySep™ magnet — the desired cells are simply poured off into a new tube and are immediately available for downstream applications such as flow cytometry, culture, and cell-based experiments.

The kit is designed for processing 1 x 10^9 cells and is reported to deliver up to 98% purity, with a typical range of 92.0–98.0% from splenocytes of an uninfected mouse. The RF (RoboSep™-S Reagent Kit) format bundles the EasySep™ Selection Kit with RoboSep™ Buffer and RoboSep™ Tip Racks for use on a fully automated RoboSep™ instrument.

Starting in early 2023, select EasySep™ Mouse kits — including this one — transitioned from including Normal Rat Serum (previously shipped separately) to including a Mouse FcR Blocker, with all components now shipped in a single package while maintaining the same isolation performance. For purity assessment by flow cytometry, STEMCELL Technologies recommends fluorochrome-conjugated antibodies against CD3e (Clone 145-2C11), CD8a (Clone 53-6.7), CD62L (Clone MEL-14), and CD44 (Clone 5035-41.1D); the IM7 clone of anti-CD44 is not recommended because it is blocked by the antibody used in the isolation cocktail.
