RoboSep Mouse Naïve CD4+ TCell Iso Kit

An immunomagnetic negative selection kit from STEMCELL Technologies enables researchers to isolate untouched and highly purified naïve CD4+ T cells (CD4+CD44lowCD62Lhigh) from mouse splenocytes. The column-free workflow uses biotinylated antibodies and magnetic particles to label and remove non-naïve CD4+ T cells, after which the desired cells are simply poured off into a new tube using an EasySep™ magnet.

Designed to be fast and easy to use, the protocol can be completed in as little as 15 minutes. Typical purity of the isolated naïve CD4+ T cell fraction (CD4+CD44lowCD62Lhigh) from mouse splenocytes ranges from 87.6–95.6%, with up to 95% purity reported. Isolated cells are immediately ready for flow cytometry, culture, or cell-based assays, and are obtained without the need for washes, supporting viability and function for downstream applications.

The kit targets non-naïve CD4+ T cells for removal using a combination of monoclonal antibodies in PBS and 0.1% BSA, together with streptavidin-coated magnetic particles. It can process up to 1 x 10^9 cells and is intended primarily for splenocyte preparations; single-cell suspensions from other tissue types may require optimization. Compatible staining markers for assessing purity include CD4, CD44, and CD62L.
