RoboSep Human EpCAM Pos Slctn Kit II

From STEMCELL Technologies, this immunomagnetic positive selection kit isolates EpCAM+ cells from fresh or previously frozen human mammary epithelial cell preparations and other dissociated tissue samples. Cells are targeted with antibody complexes recognizing EpCAM and dextran-coated magnetic particles; the cocktail also includes an antibody to human Fc receptor to minimize nonspecific binding. The EpCAM+ fraction contains luminal cells, luminal-restricted, and bipotential progenitor cells.

Labeled cells are separated using an EasySep magnet without columns—unwanted cells are poured off while cells of interest remain in the tube. Suited to cell isolation research in cancer and epithelial cell biology, the kit is compatible with the EasySep magnet, "The Big Easy" EasySep magnet, and RoboSep-S. A recommended buffer for the workflow is Hanks' Balanced Salt Solution modified with 10 mM HEPES, without phenol red.

When starting with peripheral blood mononuclear cells seeded with MCF-7 cells at a starting frequency of 10.8–50.0%, the EpCAM+ cell content (epithelial cell+CD45−) of the isolated fraction is typically 96.2 ± 3% (mean ± SD using the purple EasySep Magnet). Start and final isolated fraction purities in the same example are 10.8% and 95.0%, respectively, and purity can reach up to 99%.
