Description
From STEMCELL Technologies, this kit isolates untouched, highly purified human naïve CD4+ T cells (CD3+CD4+CD45RA+CD45RO-) from fresh or previously frozen peripheral blood mononuclear cells (PBMCs) by immunomagnetic negative selection. The fast, easy-to-use, column-free procedure achieves up to 96% purity, with the naïve CD4+ T cell content of the isolated fraction typically ranging from 91.3% to 96.9%.
The kit targets non-naïve CD4+ T cells, including regulatory T cells, for removal. Unwanted cells are labeled with Tetrameric Antibody Complexes recognizing CD8, CD14, CD16, CD19, CD20, CD25, CD36, CD56, CD61, CD66b, CD123, HLA-DR, TCRγ/δ, and glycophorin A, along with a biotinylated anti-CD45RO antibody and a bispecific Tetrameric Antibody Complex recognizing biotin and dextran. Labeled cells are separated without columns using an EasySep™ magnet, and the desired cells are simply poured off into a new tube.
The kit is designed to process 1 x 10^9 cells and is supplied with the EasySep™ Human Naïve CD4+ T Cell Isolation Cocktail, EasySep™ Biotinylated Anti-CD45RO Antibody, EasySep™ Dextran RapidSpheres™, RoboSep™ Buffer, and RoboSep™ Filter Tips for use with the RoboSep™ instrument. Components should be stored at 2 - 8°C and not frozen. Isolated cells are immediately available for downstream applications such as flow cytometry, culture, or DNA/RNA extraction.
