Description
A cell isolation kit from STEMCELL Technologies designed to enrich human CD4+CD127lowCD25+ regulatory T cells (Tregs) from PBMCs or leukapheresis samples using immunomagnetic positive selection. Formulated for use with RoboSep™ platforms, the procedure combines antibody complexes targeting CD25 with EasySep™ Releasable RapidSpheres™ magnetic particles. After separation, the bound magnetic particles are removed from the isolated CD25+ cells using a release agent, while unwanted non-Tregs are targeted for depletion.
The resulting cell fraction contains highly purified CD4+CD127lowCD25+ cells that express high levels of FOXP3 and are immediately ready for downstream applications such as functional studies, expansion, or immunological assays. The manufacturer indicates that the workflow can be completed in as little as 50 minutes when processing up to 1x 10^9 cells per isolation, with a typical CD4+CD25+FOXP3+ content of 85.0 ± 4.8% (mean ± SD) reported when starting with fresh or previously frozen PBMCs. EasySep™ technology, widely used in published research for more than 20 years, combines the specificity of monoclonal antibodies with a column-free magnetic workflow.
An optional protocol allows CD4+CD25- responder T cells to be isolated in parallel from the same sample for functional studies. The kit is intended for research use within immunology workflows.
