RoboSep Hu CD138 Pos Sel Kit II

An immunomagnetic positive selection kit in the RoboSep™ format from STEMCELL Technologies, this reagent enables the isolation of highly purified human CD138+ (syndecan-1) cells from fresh or previously frozen human bone marrow or peripheral blood mononuclear cell (MNC) samples. The system combines the specificity of monoclonal antibodies with a column-free magnetic workflow, pairing an antibody complex recognizing CD138 with magnetic particles.

In the workflow, the desired cells are labeled with these antibody complexes and magnetic particles, while the cocktail also contains an antibody to the human Fc receptor to help prevent non-specific binding. Labeled cells are separated using an EasySep™ magnet, and unwanted cells are poured or pipetted off, leaving the target cells in the tube for downstream applications such as flow cytometry, culture, or DNA/RNA extraction. The CD138 antigen is expressed on normal and malignant plasma cells, but not on mature B cells, and the underlying EasySep™ technology has been widely used in published research for more than 20 years.

The kit is intended for research use in the areas of cancer and immunology and is compatible with the EasySep™ Magnet, the "Big Easy" EasySep™ Magnet, the EasyEights™ EasySep™ Magnet, and the RoboSep™-S instrument. In a typical isolation from thawed PBMCs spiked with the U266 multiple myeloma cell line, the CD138+ content of the isolated fraction typically ranges from 93.0 - 98.2%, with start and final isolated fraction purities of 9.16% and 94.34%, respectively, in the reported profile.
