NeuroCult Chemical Dissociation, Kit

The STEMCELL Technologies chemical dissociation system enables researchers to gently dissociate neural tissue into viable single-cell suspensions for downstream neural stem cell culture workflows. This enzyme-based kit uses papain digestion combined with a DNase I treatment step to break down tissue extracellular matrix while preserving cell surface antigens and viability. The product is commonly applied to studies of embryonic or adult CNS progenitors, neurosphere assays, and in vitro differentiation protocols.

Each kit contains the papain-based dissociation enzyme, an ovomucoid protease inhibitor for terminating the enzymatic reaction, a DNase I solution, and supplemented HBSS wash buffers. The standard protocol calls for approximately 1 mL of dissociation solution per sample preparation, with an incubation period around 30 minutes at 37 degrees C to achieve optimal tissue digestion. Following dissociation, cells are collected by centrifugation and can be plated directly into serum-free NeuroCult proliferation medium or processed for flow cytometry and immunocytochemistry downstream applications.

Intended for research use only, components are shipped refrigerated and should be stored cold upon receipt to maintain enzyme activity. Researchers should aliquot working volumes of papain to avoid repeated freeze-thaw cycles, which can reduce enzymatic performance. Components remain stable for the shelf life indicated on the product label when handled according to recommended storage conditions.
