Monoclonal Anti-Human EPO-26, 100 ug

This APC-conjugated monoclonal antibody from STEMCELL Technologies, clone 5C8, recognizes human CD154 (CD40L), a 39 kDa type II transmembrane glycoprotein belonging to the tumor necrosis factor alpha (TNF-α) superfamily. The reagent is of mouse origin, was raised against the human D1.1 T cell line, and is of the IgG2a, kappa isotype. The antibody has been reported to block the interaction and ligation of CD40 with its ligand.

CD154 (CD40L) is predominantly expressed on activated T-cells and is also found on NK cells, mast cells, basophils, and eosinophils. It binds to CD40 on antigen-presenting cells such as B cells, monocytes, dendritic cells, and fibroblasts, driving B-cell activation and proliferation, antibody production, isotype switching, and B-cell memory development. CD154 expression is upregulated in inflammatory bowel disease, suggesting a role in regulating pathogenic cytokine production in T-cells, and in vitro studies show that CD40 ligation induces endothelial cell activation, consistent with an immunopathogenic role.

The antibody is verified for flow cytometry (FC). For this application, a suggested use of 5 μL per 1 × 10⁶ cells in 100 μL is recommended, with titration advised for optimal performance. It is supplied in phosphate-buffered saline (pH 7.2) containing 0.09% sodium azide and 0.1% gelatin and was purified by affinity chromatography. The product should be stored undiluted at 2 – 8 °C; it should not be frozen and should be protected from prolonged exposure to light. It remains stable until the expiry date on the label and is intended for research use only.
