Description

STEMCELL Technologies supplies a mouse monoclonal antibody (clone AA10, IgG2a kappa) raised against human, mouse, and rat beta-tubulin III, an approximately 50–55 kDa structural protein that forms part of the microtubule cytoskeleton. The beta III isoform, also referred to as neuron-specific class III beta-tubulin, is expressed primarily in neurons and is widely used as a marker to distinguish neurons from other cell types. The antibody contributes to studies of microtubule formation, axonal transport, neuronal proliferation, and differentiation, and is expected to recognize all mammalian homologs of beta-tubulin III.

The reagent is provided unconjugated in a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide, and has been purified by affinity chromatography. It was generated using a synthetic peptide corresponding to amino acids 436–450 of beta-tubulin III conjugated to keyhole limpet hemocyanin. Verified applications include immunocytochemistry (ICC), immunofluorescence microscopy (IF), and Western blotting (WB), with reported use also in immunohistochemistry (IHC) and immunoprecipitation (IP). Suggested working concentrations are ≤ 4 μg/mL for ICC/IF and ≤ 2 μg/mL for WB, with titration recommended for each application. The product is stable at 2–8 °C when stored undiluted and should not be frozen.

The clone has been verified for labeling neural stem and progenitor cells cultured with STEMCELL Technologies' Neural Induction Medium, Neural Progenitor Medium, and NeuroCult™ proliferation and neuronal culture kits. Beta-tubulin III is also recognized as a predictive and prognostic marker in several tumor types, being found in neoplastic but not in normal glial cells.
