Description

This mouse monoclonal IgG2a, kappa antibody from STEMCELL Technologies targets β-tubulin III, a ~50 - 55 kDa structural protein that is a major constituent of microtubules within the cytoskeleton. The AA10 clone reacts with the β III isoform of tubulin, also known as neuron-specific class III β-tubulin (TUBB3), which is expressed primarily in neurons and is widely used as a marker to distinguish neurons from other cell types. The protein contributes to microtubule formation in neuronal cell bodies and axons, a function involving GTP binding, and plays roles in axonal transport, neuronal cell proliferation, and differentiation. It is also highly expressed in several types of cancer and serves as a predictive and prognostic marker for various tumors, having been reported in neoplastic but not in normal glial cells. The antibody is expected to recognize all mammalian homologs of β-tubulin III.

The reagent is conjugated with Alexa Fluor® 488 and shows reactivity with human, mouse, and rat β-tubulin III. Verified applications include immunocytochemistry (ICC) and immunofluorescence (IF); flow cytometry (FC) has been reported in the literature. For immunocytochemistry, the suggested working concentration is ≤ 1.5 μg/mL. The antibody was raised against a synthetic peptide corresponding to amino acids 436 - 450 of β-tubulin III conjugated to keyhole limpet hemocyanin, then purified by affinity chromatography and conjugated with Alexa Fluor® 488 under optimal conditions; the solution is free of unconjugated Alexa Fluor® 488. It is supplied as a phosphate-buffered solution at pH 7.2 containing 0.09% sodium azide. The product is stable at 2 - 8 °C when stored undiluted; it should not be frozen and should be protected from prolonged exposure to light.
