ImmunoCult NK Cell Expansion Kit

STEMCELL Technologies' ImmunoCult NK Cell Expansion Kit is a culture medium system developed for the in vitro culture and expansion of human natural killer (NK) cells. The kit comprises three components: a serum-free and xeno-free base medium containing recombinant and plasma-derived proteins and synthetic components (growth factors are not included, allowing users flexibility to supplement the medium as required); an animal component-free expansion supplement supplied as a 100X concentrate, containing recombinant human cytokines formulated to support NK cell expansion; and an animal component-free coating material supplied as a 100X concentrate, formulated to promote NK cell expansion when applied to non-tissue culture-treated cultureware.

The workflow supports expansion of NK cells from peripheral blood mononuclear cells (PBMCs), or from NK cells isolated from fresh whole blood or leukapheresis samples, with cultures incubated at 37°C and 5% CO2. Cultureware is coated with the diluted coating material for 2 hours at room temperature (15 - 25°C) or overnight at 2 - 8°C. The kit has also been demonstrated in CRISPR-Cas9 genome editing protocols, where primary human NK cells are expanded for 3 - 4 days on pre-coated plates prior to delivery of a Cas9 ribonucleoprotein (RNP) complex by electroporation using either the Neon® Transfection System or the Lonza® 4D-Nucleofector™ X Unit. Compatible NK cell isolation reagents include the EasySep™ Human NK Cell Isolation Kit and EasySep™ Human NK Cell Enrichment Kit.
