ImmunoCult Mouse B Cell Expansion Kit

A serum-free, feeder cell-free culture kit from STEMCELL Technologies, developed for the in vitro expansion of mouse splenic B cells. By combining the base medium with the expansion supplement, the system supports consistent activation and expansion over 9 days of culture, with cells suitable for downstream applications after 6 - 9 days. The supplement is animal component-free and supplied as a 50X concentrate.

B cells are isolated from freshly processed mouse splenocytes using the EasySep Mouse Pan-B Cell Isolation Kit; this kit does not support thawed frozen mouse pan-B cells. Cells are seeded at 1 - 2.5 x 10^5 cells/mL and incubated at 37°C and 5% CO2, with passaging on days 3 and 6. Under these conditions, B cells expanded 176.9 ± 29.8-fold after 9 days of culture (mean ± SEM, n = 8). Expansion is accompanied by surface-marker changes typical of B cell activation and maturation to plasmablasts or plasma cells, including upregulation of CD86 and CD138, downregulation of B220, and increased TACI (CD267) expression.

Storage requirements differ by component: the base medium is stored at 2 - 8°C and remains stable until the expiry date on the label, while the supplement is stored at -20°C and is stable for 3 years from the date of manufacture. The base medium contains material derived from human plasma and should be handled using universal precautions. For research use only; not intended for human or animal diagnostic or therapeutic uses.
