ImmunoCult Human B Cell Expansion Kit

The STEMCELL Technologies ImmunoCult™ Human B Cell Expansion Kit is designed to support consistent expansion of human B cells without the use of serum, specialized cultureware, or feeder cells. The kit combines two components: ImmunoCult™-ACF Human B Cell Expansion Supplement, a serum-free and animal component-free culture supplement, and ImmunoCult™-XF B Cell Base Medium, a serum-free and xeno-free base medium for B cell expansion. Together they enable activation, expansion, and maturation of human B cells toward plasma cells and blasts for downstream applications.

According to manufacturer data, B cells isolated from human PBMCs and seeded at 1 x 10^5 cells/well in 24-well tissue culture plates with the kit achieved a fold expansion ranging from 38- to 1190-fold at day 14 ± 1 day across n = 12 donors, with passaging every 3–4 days. Flow cytometry analysis of CD138 and CD20 expression at each timepoint indicated maturation of B cells to plasma cells and blasts. The kit has also been shown to expand memory B cells and naive B cells seeded at 0.5 x 10^5 cells/well in 48-well plates, with passaging every 2–4 days.

Following isolation with EasySep™ cell separation kits for pan-B cells, memory B cells, or naive B cells, the isolated subsets can be expanded directly with this kit, fitting into upstream and downstream immunology workflows.
