Description

A culture medium kit from STEMCELL Technologies provides three complete cell culture media designed to support the initiation, efficient long-term expansion, and differentiation of human liver organoids derived from ductal stem cell niches isolated from primary human liver tissue. The three included media are also available as individual kits for organoid initiation, organoid growth, and organoid differentiation.

Hepatic organoids generated with this system offer a three-dimensional in vitro organotypic culture system for the study of hepatic development, regeneration, detoxification, metabolism, and disease. Organoids initiated in the Organoid Initiation Medium and expanded in the Organoid Growth Medium express self-renewal markers (LGR5, AXIN2), ductal markers (KRT19, SOX9, KRT7), and the hepatic progenitor marker HNF4A. Upon switching to the Organoid Differentiation Medium, the organoids form mature hepatic structures that express hepatocyte markers (ALB, CYP3A4, MRP4) and exhibit upregulated albumin secretion and CYP3A4 activity relative to undifferentiated organoids.

Organoids expanded in the Organoid Growth Medium can be maintained in Corning® Matrigel® domes for routine culture, in dilute Matrigel® suspensions in spinner flasks for scale-up, in 96- and 384-well plates for high-throughput screening, and as organoid-derived two-dimensional monolayers on Transwell® inserts for disease modeling. The organoids can also be cryopreserved for later use, remaining highly viable and retaining capacity for further differentiation when thawed and cultured in the Organoid Growth Medium. The product was developed under license to intellectual property owned by Hubrecht Organoid Technology (HUB) and is sold for research use only.
