EasySepHLA WB Ttl Lymphocyte Enrich Kit

STEMCELL Technologies offers an immunomagnetic negative selection kit for isolating highly purified, untouched human total lymphocytes directly from HetaSep™-treated whole blood or washed leukapheresis samples. The column-free procedure targets non-lymphocyte cells for removal using antibodies against cell surface markers including CD14, CD15, CD16, CD33, CD36, CD41, CD66b, and glycophorin A, combined with tetrameric antibody complexes and dextran-coated magnetic particles that are separated without columns using an EasySep™ magnet. Desired cells are simply poured off into a new tube.

The kit is fast and easy to use, delivering up to 97% purity with untouched, viable cells that are immediately compatible with downstream HLA assays, flow cytometric crossmatch analysis, culture, and DNA/RNA extraction. Starting with whole blood, the lymphocyte (CD3+ and CD19+) content of the enriched fraction is typically 96.8 ± 0.5% (mean SD using the purple EasySep™ Magnet), and the CD3+ / CD19+ cell content of the enriched fraction typically ranges from 90.2 - 96.9% (gated on CD45+ cells). Starting with 0.5 mL of HetaSep™-treated whole blood (equivalent to 5 mL of whole blood), 1.3 - 3.6 x 10e6 CD3+ or CD19+ cells are typically recovered.

Components include an Enrichment Cocktail (a combination of monoclonal antibodies in PBS), EasySep™ D Magnetic Particles (a suspension of magnetic particles in TBS), and HetaSep™ 6% Hetastarch Solution used for erythrocyte aggregation. Compatible magnets include the EasySep™ magnet, "The Big Easy", EasyEights™, Easy 50, and RoboSep™ instruments.
