STEMCELL Technologies offers this immunomagnetic kit for isolating highly purified human CD3 and CD19 lymphocytes from fresh human whole blood, buffy coat, or an LRS cone for chimerism analysis following HSC or bone marrow transplantation. The procedure uses column-free magnetic separation on the EasySep™ platform, with target cells labeled by antibodies recognizing CD3 and CD19 together with Tetrameric Antibody Complexes that bind dextran-coated magnetic particles. The antibody cocktail also includes reagents that recognize human Fc receptors, reducing non-specific binding. Unwanted cells are removed simply by pouring off the solution, while the magnetically labeled target cells remain in the tube.

When starting with human whole blood, the lymphoid cell content (CD3+CD19+) of the isolated fraction is typically 99.5 ± 0.2% (gated on CD45; mean ± SD using "The Big Easy" EasySep™ Magnet), with the purity of the starting and final isolated fractions reported as 16.2% and 98.5%, respectively. The CD3 antigen is expressed on T lymphocytes and NKT cells, while the CD19 antigen is expressed on B cells and follicular dendritic cells.
